18 16 / Accompanying Conference According to the ISO definition, quality assurance involves all the planned and systematic activities implemented within the quality system and demonstrated as needed, to provide adequate confidence that an entity will fulfil requirements for quality. In plain English, quality assurance is what the analysts do to make sure that the answers they produce are correct. No analytical measurement is perfectly accurate, and it is the role of the analyst to minimize the uncertainty. Before performing any measurements, the objectives and the specifications should be defined. Quality assurance itself begins with proper sampling. Sample chain of custody should be properly documented. Any method used for the analysis should be properly validated by determining a number of characteristics including specificity, linearity, accuracy, precision, range, limits of detection/ quantitation, as well as reporting limits. Robustness of the method should also be evaluated. Once a validated method has been implemented, its performance should be monitored continuously. Analysis of blanks makes it possible to account for interferences by other species and for traces of the analyte that may be present in the reagents used for sampling, sample preparation and analysis. Various types of blanks should be analyzed, including reagent blanks, method blanks, trip blanks, field blanks, etc. Potential matrix effects should be evaluated by analyzing matrix spikes. During the final determination step, stability of the instrument calibration should be closely monitored by including calibration checks at regular intervals. The correctness of the calibration itself is monitored by periodically analyzing performance test samples. All steps involved in quality assurance should be described in detail in standard operating procedures that all personnel involved in the analysis must adhere to. These figures of merit are usually associated with the analyzer. Driving these figures of merit to hitherto unheard levels, novel developments include the Orbitrap analyzer commercially introduced in 2005 and still evolving, ion funnels and ion transfer stages in various novel powerful designs, differential mobility devices such as FAIMs, to name a few. Along this line, more than 50 new acronyms for novel ion sources operating at atmospheric/ambient pressure have been coined in the past two decades suggesting a parallel development with regard to modern ion sources, including DART, DESI, and many, many more. Nevertheless, ions generally formed in the various API sources are mostly protonated molecules [M+H]+, sometimes adducts [M+X]+, and, to a far lesser extent, radical cations M +. This suggests that at least comparable ionization mechanisms are generally operative in API despite the numerous methods developed so far. The question arises, why is that? In order to unravel the various deeply folded processes operative in API sources, we will first have a look at the classical methods i) electron ionization (EI), and ii) chemical ionization (CI), where the latter is driven by EI of reagent gases, generating the CI reagent ions, which in turn ionize the analyte(s) present. The typically collision free environment in EI leads to very well documented and compiled ion populations comprised of molecular radical and fragment ions, respectively. Even theories such as Quasi Equilibrium Theory (QET) for the molecular description and calculation of ionic fragmentation pathways were successfully developed and the predictions are quite reasonable. A similar picture can be drawn for classical CI, i.e. operation at source pressures around 1 mbar with the source matrix gas comprised essentially of the reagent gas: The generated excess reagent ion population usually drives all chemical equilibria involving analyte ionization steps to completion. In other words: Thermodynamical properties such as gas phase acidities and basicities can be taken with some confidence to predict the generated analyte ion population. This picture largely changes in API. Here, phenomena as for example i) ion suppression, ii) efficient formation of protonated molecules despite the usage of dry nitrogen as discharge gas, iii) shifts towards higher analyte charge states upon super charging agents to the gas matrix in nanoelectrospray, and iv) the frequent failure when using thermodynamical properties such as gas phase acidities/basicities for the prediction of AP ionization mechanisms, are repeatedly observed. Again, the question arises, why is that? These issues and phenomena will be highlighted in some depth in the second part of the tutorial. Ion cluster chemistry will play a pivotal role. It will be illustrated how cluster ion chemistry potentially drives ionization pathways into unexpected directions along with a discussion on the frequent absence of cluster ions in the recorded API mass spectra.

Novel developments in high resolution mass spectrometry (HRMS) or significant improvements in sensitivity and speed of MS instrumentation now allow for rapid multiplexing as well as in-depth analysis of complex food samples. Food proteomics is such an emerging novel field in food analysis that is driven by developments in MS instrumentation. In this tutorial, we will exemplify novel approaches to investigate authenticity and safety of food using food proteomics. The identification of biomarker peptides for authenticity or the detection of food allergen trace contaminations is discussed as well as possibilities for the detailed characterization of bioactive food proteins (allergens, celiac disease). From the methodological point of view, the potential and pitfalls of targeted and untargeted methods is discussed as well as the use of novel MS setups such as MRM3. The use of these techniques for non-protein analytes is also a topic of this tutorial. 20 18 / Accompanying Conference Solventless Extraction is an environmentally friendly technique used in sample preparation. It has developed rapidly in recent years and is preferred in laboratories and industry due to its time, labor and cost-savings and in particular its elimination of solvent and minimization of sample size. High recovery, sensitivity, and low detection limits are routinely achieved. In the first part of this tutorial, several solventless extraction techniques will be introduced, such as headspace (HS) analysis, solid phase microextraction (SPME), stir bar sorptive extraction (SBSE), chamber emission testing and pyrolysis. In the next two presentations, dynamic headspace (DHS) techniques and thermal desorption (TD) techniques will be presented in detail with respect to their usability and effectiveness for selected samples in the fields of food, flavor and fragrance (FFF) or material testing and characterization. In the last presentation, recent developments in automated microextraction techniques will be presented by Prof. Ouyang from Sun Yat-sen University. Extending Dynamic Headspace (DHS): Dr. Xiao Li, GERSTEL LLP, Singapore (language: Mandarin) Two ways of extending the power of DHS, the full evaporation technique (FET) and multi volatile methods (MVM) will be introduced. By fully evaporating the solvent matrix, FET can extend the volatility range of compounds compared to standard HS, DHS, or SPME. MVM uses sequential DHS sampling to analyze VVOCs, VOCs and SVOCs from aqueous samples and can cover an even wider range of volatility of compounds (vapor pressures from ~ to 120 kpa). The performance of DHS, FET and MVM will be demonstrated using aroma compounds in tea, brewed coffee, shampoo and milk powder at sub-ppb to ppm levels. TD Related Techniques: Dr. Eike Kleine-Benne, GERSTEL GmbH & Co.KG, Germany (language: English) Thermal Desorption (TD), a sample extraction technique and sample introduction technique for gas chromatography is discussed with a focus on material characterization. Some application examples are screening interior materials (e.g. from vehicles or buildings) for VOC and SVOC emissions to direct thermal extraction of particulates captured on PM 2.5 dust filters. Fully automated micro-scale emission chamber screening methods to determine VOC and SVOC emissions from building and insulation materials will be considered. Finally, pyrolysis for the characterization of polymers and nonvolatile compounds will be reviewed. In this study, fully-automated microextraction techniques, including headspace solid-phase microextraction (HS- SPME), coupled with GC/MS and HPLC were investigated with the use of Gerstel Multi Purpose autosampler, sample extraction, injection and analysis procedures can be automatically carried out.
