Leica EM GP 样 品 快 速 冷 冻 仪 快 速 操 作 流 程
1. 把乙烷杯和样品盒转移容器放入液氮杜瓦 并注入液氮冷却 2. 开机 轻触屏幕后 出现工作界面
3. 设置样品腔室温度 TC 湿度 HR 乙烷温度 4. 放上乙烷加注罩 加注乙烷
5. 注入 60ml 的去离子水 6. 更换滤纸 7. 进行制样参数设置
下一步 8. 用镊子加上载网安装在杆上 9. 点一下 Lower Chamber /R,把样品腔降下来
10. 在载网上加上样品 11. 点 Rotate Home Blot S /A- Plunge,
12. 点 Transfer
13. 用手把镊子从杆上取下 14. 把冻好的样品转移到样品盒中 15. 冻完样品 用盖子把样品转移盒盖上 用专用螺丝刀把样品转移 盒从杜瓦中取出放入提前准备好的容器中
16. 用完仪器 点 Bakeout 17. 等温度升到室温后 关机即可
Overview of Leica EM GP 1
Leica EM GP operation 1. Preparation Turn on the power and touch screen to start; Position the black secondary cryogen container in its holder; Load the marked grid box into the transfer (TF) container; Position TF container on its platform in the Dewar chamber; Cryogen container TF container Grid box Take a filter paper ring and place the metal ring over it, hold them together and insert into the magnetic mount in the environment chamber; Pour LN2 directly into the Dewar chamber until the shows 100%; Note that you can also fill some LN2 into cryogen container and TF container to cool down; Add 60ml distilled water via the silicon tube on the right hand side of the environmental chamber by filling a syringe, locking it onto the tube with the Luer lock and dispensing ; CLosed Open Fill ethane into the cryogen container with ethane liquefier until the container is full. Note that the cryogen valve should be opened slowly to avoid spilling ethane out. 2
2. Parameters settings Touch any part of the setting panel of the screen will open the corresponding setting window. chamber environment settings Open the left panel, set the chamber temperature (Tc) and humidity (HR) and ethane temperature. program settings Touch and open the program parameters. For single blot: Rotate grid before applying Program sample number Blotting with blot sensor actived Plunge automatically after blotting Blotting time Rotate grid after applying sample Pre-blot time before blotting Post-blot time before freezing after blotting Choose a program, check the first four boxes to activate them, and set the pre-blot, blot and post-blot time in sequence. Good blotting time for most suspensions of macromolecules are 0.7 sec to 3.5 sec. Press ok to return to the main screen. Notes: By ticking auto plunge, after Blot and post-blot, the grid continues automatically to plunge into the ethane. The plunge can be made manually when the autoplunge box is not ticked. In this case the button has to be pressed after blotting. 3
For multiple blot: Blotting with blot sensor activated Enable and box, set suitable pre-blot, blot and post-blot time, set times. Notes: this multiple-blot setting means that this program will blot 3 times at grid home position and 2 times at grid right port position. By ticking the grid will rotate back after adding the fluid. So the alternate blotting starts on the grid home position (Coating/sample side to the left). If the box is not ticked, blotting starts on the grid right port position. Adjust grid /blotter positions Press to show the setup main menu. First, press in blotter settings frame you can get the popup window, then press Blot Position button and with the button the blotter can be moved left and right to bring filter paper and grid into the perfect juxtaposition. 4
To test the blot, press then the length of time of the test blot can be adjusted by pressing on And press Back to return to the setup menu. Second, press in the Grid Blot Position frame, then press on the pop-up window, press the blotter can be raised or lowered to the desired position. And press Back to return to the setup menu. Third, in order to remove easily, transfer position can also be adjusted. Press Adjust in Grid TF Position frame, then press Then by pressing the adjust arrows, the grid height can be set. Pressing takes the grid to the set point. Press to go back to the main screen. 3. Run the program After all parameters is well set, you can run the programs. Press to raise the environment chamber to the upper position. pick up a grid with forceps, lock it in place with the locking sleeve and push the forceps into position on the gantry of the EM GP. Note that the carbon side of grid should be pick correctly and Home position and Right port position should be differentiated. Carbon side Home position Right port position 5
Press the chamber will move down to cover the grid and forceps. Add 4µl fluid sample onto the grid, close the sliding pipette access door. Press your specific program will run. The countdown box will show the progress. After the program finished, forceps will plunge into ethane immediately if you check the box, or you can press to plunge manually. Transfer the grid to the grid box. And the next run can be started. 4. Closing down after cryo fixation On completion of the cryowork the EM GP must be warmed to drive off any moisture which may cause problems on the next use by freezing up moving parts. Press Bake out and open the environmental chamber door Press and remove cryogen container and TF container in the fume hood. Press Start Bake Out. Open the chamber door, and clean up the water reservoir. If the chamber contains LN2, bake out cycle will not start. Therefore, complete bake out time depends on LN2 level and environment temperature. Turn off the power. 6
