RNA 提取 1) 取 0.25 ml RNAzol RT RNA 抽提试剂加入装有 50 μl 或 100 μl 澄清粗慢病毒溶液 ( 或 10 μl 纯化浓缩的慢病毒溶液 ) 的 1.5 ml 离心管中, 将离心管颠倒 10 次混匀溶液 ( 裂解病毒颗粒并溶解蛋白 ), 室温静置至少 15 min 2) 短暂离心后, 根据步骤 1.1) 中慢病毒溶液添加量, 参考下表向步骤 1.1) 加入一定量纯水 慢病毒 100 μl 粗慢病毒 50 μl 粗慢病毒 10 μl 纯化慢病毒 水 0 50 μl 90 μl * 100 μl Note: 表格中 100 μl 体积为慢病毒与水的总量, 不包括 RNAzol RT RNA 抽提试剂 3) 匀浆液在 20 C 条件下 18,000 g 离心 10 min 4) 小心地将上清液转移至新的 1.5 ml 离心管中, 加入线性聚丙烯酰胺 (Linear Polyacrylamide), 使其终浓度约为 15 μg/ml 例如, 在 100 μl 上清液中加入浓度为 1.5 mg/ml 的线性聚丙烯酰胺 1.0 μl 注意! 线性聚丙烯酰胺作为共沉淀剂, 可以改善乙醇沉淀过程中 RNA 的恢复, 也有助于 RNA 沉淀的显现, 但不会影响后续的酶消化 反转录和 qpcr 反应 5) 加入上述溶液相同体积的 100% 异丙醇 ( 或 3 倍体积 100% 乙醇 ), 反复颠倒离心管混匀溶液 混匀的溶液最好在 -20 C 下保存 4 小时以上或者过夜 6) 溶液在 10 C 条件下 18,000 g 离心 20 min, 并弃去上清液 7) 用 0.5 ml 75% 乙醇洗涤 RNA 沉淀, 溶液在 10 C 条件下 18,000 g 离心 5 min, 弃去上清液 再重复洗涤过程一次 8) 尽可能除去残留乙醇 室温下晾干 RNA 沉淀 3 min, 切勿过干 最后用 50 μl 的 DEPC Water 溶解 RNA 沉淀 2
2. 用 DNase I 处理 ( 去除游离细胞基因组及包装质粒 ) DNase I 反应 用 1.5mL 管子进行以下反应 ( 总体积 25 μl): DEPC water Lentiviral RNA DNase I buffer (10 ) DNase I 1.5 μl 20.0 μl 2.5 μl 1.0 μl 25.0 μl 孵育 : 137 C, 30-60 分钟 275 C, 10 分钟 ( 使 DNase I 失活 ) Note: 如果遗漏了 DNase I 消化步骤, 必须在 qpcr 反应步骤中加上以未反转录的 RNA 样品为模板的 qpcr 反应作为对照, 该对照确定 ( 未经过 DNase I 消化的 ) 样品中所携带的质粒 DNA 拷贝数, 用反转录产物作为模板进行的 qpcr 反应所确定的拷贝数减去该对照确定的质粒 DNA 拷贝数即为样品中 RNA 拷贝数 3. 反转录 1) 在 0.2 ml 或 0.5 ml 离心管中制备 RNA-Primer Mix, 混匀 RNA-Primer Mix, 在 70 C 温育 5 min 后, 将离心管立即置于冰上至冷却 RNA( 经过 DNase I 消化过的 ) cdna Synthesis Primer(4.0 μm) 10.0 μl 5.0 μl( 终浓度 1.0 μm) Note: 随机引物 ( 在反转录反应液中的终浓度为 10 μm) 可用于替代 HIV cdna Synthesis Primer 无需同时使用 cdna Synthesis Primer 和随机引物 2) 准备反转录反应体系, 继续加入其它组分 ( 总体积 20 μl), 短暂离心 ( 混匀反应液并富集于离心管底 ),37 C 温育 60 min Reverse Transcription Buffer (10 ) 25 mm dntp 1.0 μl RNase Inhibitor Reverse Transcription Enzyme 1.0 μl 1.0 μl 20.0 μl 3) 90,10 min 该产物可直接用于 qpcr 检测实验, 或者保存于 -20 C 4. qpcr 反应 1) 准备制作标准曲线样品稀释阳性标准品制作标准曲线 ( 每个稀释梯度取 5 μl 作为模板进行 qpcr 反应 ) (1) 2 x 10 7 copies/μl (5 μl of qpcr standard (DNA) + 245 μl of ddh2o) (2) 2 x 10 6 copies/μl (5 μl of (1) + 45 μl of ddh2o) (3) 2 x 10 5 copies/μl (5 μl of (2) + 45 μl of ddh 2O) 3
