Methods The modified gradient procedure was used to separate the lactose standard on a CarboPac PA20 (3 mm 150 mm) anion exchange column which was established automatic analysis method and used ion chromatography and external standard methods for quantitative analysis. Results Under optimal experimental condition, the lactose linear range of 0.2~20 mg/l, the correlation coefficient was greater than 0.99. The method detection limit (S/N=3) was 10 mg/kg, the quantitative limit (S/N=10) was 30 mg/kg; lactose of human milk value was 0.72%~1.53%. Conclusion After method optimization, excluding impurities interference, lactose was determined more accurately. This method operation is simple, accurate and cheap, suitable for lactose in human milk analysis of the test.
