Abstract: Background: The development of autoantibodies in the sera of patients with cancers has been described, but the mechanism is still not clear. Objective: The aim of this study was to analyze the autoantibodies in the serum of cancers to further understand the relationship among the autoantibodies and between autoantibodies and cancers. Methods: Sera were collected from patients with cancers without autoimmune disease, including HCC(hepatocellular carcinoma), GC(gastric cancer), and the cancer of lung, rectum, and esophagus.recombinant p62, Ro/SSA(52) and Ro/SSA(60) antigen were used to detect autoantibodies in serum from patients with HCC, GC patients with cancer of lung, rectum, and esophagus by ELISA. Immunofluorescence and Western blot were used to characterize the autoantibodies in the patients with HCC and GC. Result: We found that autoantibodies which react with cancer-associated-antigens are extensively present in sera of patients with HCC and GC. Among patients with cancer, the class of IgM,IgG and IgA of anti-p62 anti-ro/ssa(52) and anti-ro/ssa(60) was detected, positive rate ranging from 1.8% to 43.6%. It is revealed in the study that the anti-p62 IgG autoantibodies in the patients with GC compared to controls(p<0.05),and the patients with cancer of lung and rectum(p<0.01). Except the anti-ro/ssa(60) autoantibody in sera from patients with GC and esophagus, significant difference is observed between patients and controls. There is a significant difference between anti-ro/ssa (52) and anti-ro/ssa (60) autoantibodies in patients with cancer of lung and controls. In contrast, no significant difference between the male and female patients was detected. Conclusion: Diversity was found in the autoantibodies in the HCC and GC. These results indicate that p62 is an antigen of cancer, which can serve as a marker of malignancy in the cancer of lung, hepatocellular, and rectum. This is believed to be the first report showing anti-ro/ssa is associated with cancer of esophagus, gaster, rectum, hepar and lung. All these results reflect the important role of the autoantibodies in the patients with cancers. Key word: p62, Ro/SSA(52)/Ro/SSA(60), autoantibody, cancer
