16 引物的长度通常为 17~30 个核苷酸如 8nt 的引物, 平均每 4 8 =65536 bp 就会有一个结合位点, 在全长为 bp 的人类基因组中, 大约有个可能的结合位点, 不能得到单一的特异性扩增产物如为 17nt 的引物, 出现几率为 4 17 =17,179,869,184 bp, 长度超过人类基因组长度的 5 倍, 故在人类基因组中只可能有一个结合位点但引物不是越长越好, 过长的引物同模板 DNA 的结合效率反而下降, 降低 PCR 反应的效率 16

21 引物与复性温度复性温度常与引物的长度和碱基组成有直接关系, 引物越长或 GC 含量较高, 退火的温度可以高些, 反之则低些 Tm=4 (G+C)+2 (A+T) ~5 below 引物与模板结合的特异性与复性温度密切相关当复性温度偏低时, 引物与模板配对出现错配碱基, 导致一些不需要的 DNA 片段被扩增复性温度过高, 引物与模板不能配对 21

49 8. Hot-start PCR One of the most frequently encountered problems is the annealing of the primers at the wrong locations to generate the wrong PCR products. DNA polymerase is added when the reaction tubes have reached the DNA melting temperature of the first cycle. Incorporate the polymerase or the magnesium salt into wax beads or have the polymerase inactivated at the start by complexing with an antibody. 49

50 9. Touch-down PCR A high annealing temperature is used initially to ensure the correctly matched primertemplate annealing. The annealing temperature is then reduced in subsequent rounds to efficiently amplify the target sequence. 50

58 Absolute quantitation Using competitive RT PCR, measures the absolute amount (e.g copies) of a specific mrna sequence within a sample. Dilutions of a synthetic RNA (containing identical primer binding sites, but slightly shorter than the target RNA) are added to the sample and are co-amplified with the target. The PCR product from the endogenous transcript is then compared with the concentration curve created by the synthetic competitor RNA. 58
