Main research area: 1) Layer-by-layer self-assembly and biomimetic interfaces. 2) Biomaterials for cardiovascular implants and devices. 3) Bionanointerfaces. 4) Smart biomimetic polymeric nano-carrier. 1) Cardiovascualr Biomaterials: The importance of the EC competition over SMC for in-situ endothelialisation of a cardiovascular stent Cardiovascular stent implantation has been the most important procedure in the treatment of cardiovascular diseases. The in vivo endothelialisation of cardiovascular stent provides a promising strategy for in-stent restenosis and thrombosis. Based on the 5 years work in three independent approaches, we recently demonstrated that the efforts to partially enhance the endothelial cells(ecs) growth without considering the cell competition might be misleading and meaningless in vivo. Instead of the rapid endothelialisation, the competitive ability of ECs over smooth muscle cells (SMCs) is a 5 P a g e
significantly more important criterion for the development of a pure endothelial layer in vivo and thus the attainment of a better anti-restenosis effect. Based on those technique, three kinds of cardiovascular stent have been developed and in clinic trail stage. 2) Bionanointerface: from zwitterionic ligand to mixed charge A great promise for nanotechnology in medical applications has focused on the development of different forms of nanoparticles for cancer therapy. However, for further biomedical applications, these nanoparticles should not only be high colloidal stable and biocompatible in the physiological environment, but also present targeting to tumor cells. Compared to the normal PEG based modification approaches, we find the small molecular of zwitterionic ligands and mixed charge ligands present more interesting properties for surface tailoring of nanoparticles with high dispersible stability, better stealth properties and unique tumor targeting properties. The covalent conjugation of PC onto GNs not only brings better dispersion stability than the neutral EG4, but also leads to targeting cell uptake within cancer cells. We further demonstrated that the surface tailoring of nanoparticles via mixed-charged SAM can provide a facile method to present better stealth properties and higher accumulation in tumor than PEG-2000 modified nanoparticles. Combing with the ph-responsive properties of weak electrolytes, the mixed charge bionanointerface can be explored as a robust method to precisely control the aggregation of NPs to enhance the retention and cellular uptake of inorganic NPs in tumors, which has perfect stealth properties and ph-sensitivity for tumor targeting and photothermal therapy. 3) Smart Drug Delivery Systems) Stimuli-responsive polymeric assemblies, including micelles and vesicles, have attracted considerable attention due to its great potential in the field of smart and targeted drug delivery. Several smart drug delivery systems, including ph-responsive DDS, photo-responsive DDS etc, has been developed in our lab.
