1 一實驗原理 1.1 穿透式電子顯微鏡構造穿透式電子顯微鏡分析技術穿透式電子顯微鏡 (Transmission Electron Microscope) 的構造可分為光學系統真空系統和高壓電源控制系統三大部分光學系統是穿透式電子顯微鏡的主體, 其構造如圖 1.1, 包括電子槍電磁透鏡像差補償器螢幕照相機等, 各部位之功能敘述如下 : 1. 電子槍 (Electron Gun): 由高壓電源控制系統提供高電壓 (~200kV), 產生電子束當做光源 2. 陽極 (Anode): 吸引電子槍之熱電子形成電子束, 並且接地 3. 第一聚光透鏡 (Condenser 1): 控制電子束大小 (Spot Size) 4. 第二聚光透鏡 (Condenser 2): 控制輝度 (Illumination) 5. 聚束光欄 (Condenser Aperture): 控制電子的角度, 並除去高角度電子 6. 物鏡 (Objective Lens): 聚焦 7. 像差補償器 (Stigmator): 調整電磁透鏡之磁場, 用以校正散光像差 (Astigmatism) 8. 第一中間透鏡 (Intermediate Lens 1): 調整影像 (Image) 和繞射 (Diffraction) 的放大倍率 9. 第二中間透鏡 (Intermediate Lens 2): 用以切換影像或繞射 10. 投影透鏡 (Projector Lens): 控制最後之放大倍率 11. 快門 (Shutter) 螢幕 (Screen) 曝光計 (Exposure Meter) 相機 (Camera) 1.2 電子槍電子槍為電子顯微鏡的光源, 電子槍包括燈絲 ( 陰極 ) 做為電子源和加速電子之陽極, 可分為熱游離電子槍 (Thermionic Electron Gun) 和場發射電子槍 (Field Emission Gun) 兩種, 分別介紹如下 : 熱游離電子槍大部分的穿透式電子顯微鏡使用此型電子槍, 其構造如圖 1.2 當燈絲被加熱到白熱化時即放出熱電子 (Thermionic Electrons), 熱電子在 Wehnelt 圓柱體 (Wehnelt Cylinder) 孔正下方附近節縮成最小斷面積,C 點稱為電子束最小交迭點 (Cross-over Point) 熱電子被施加於陽極的高電壓 (~200kV) 加速而進入第一聚光透鏡對於電子槍, 我們可藉由以下三個參數而加以控制 :(I) 燈絲電流 (Filament Current),(2) Wehnelt 圓柱體偏壓 (Wehlelt Cylinder Bias),(3) 加速電壓 (Accelerating Voltage) 1.2.1a 燈絲電流燈絲電流的大小決定燈絲溫度的高低而影響熱電子的放射量隨著電流增加, 熱電子的放射量也愈多, 熱電子流也愈大最後, 熱電子的放射量達到飽和 (Saturation), 即使燈絲溫度繼續上升, 熱電子的放射量亦不變如圖 1.3 所示燈絲電流的控制相當重要, 因為它直接影響燈絲的壽命

2 1.2.1b Wehnelt 圓柱盤偏壓 Wehnelt 圓柱體偏壓控制熱電子流的大小和燈絲上可放射熱電子的面積之大小降低偏壓可減少電子束所造成的損害, 增加偏壓則可觀察較厚的試片 1.2.1c 加速電壓當提升工作電壓時, 是從低壓的 20kV 或 40kV 漸漸升至 200kV( 視機型而定 ) 加速電壓的選擇是由試片的種類和厚度決定電壓愈高, 電子的能量愈高, 可穿透較厚的試片, 缺點是會使得對比 (Contrast) 降低常用的熱游離電子槍有兩種, 燈絲分別為 V 字形的鎢 (W) 和針尖狀的六硼化鑭 (LaB 6 ), 如圖 1.4 所示燈絲需加熱至高溫, 電子才能獲得足夠的能量而脫離出來, 如圖 1.5 所示其電流密度 J c 與發射溫度 T 及功函數 f 之關係如 Richardson 定律表示 : J c =A c T 2 exp(-φ/kt) (A/cm 2 sr) A c 是常數與材料有關鎢的功函數是 4.6eV,LaB 6 是 2.4eV 熱游離電子槍的缺點是電流低電子束面積大輝度小另一種熱游離式的 Schottky Emmiter 電子槍, 鎢針表面鍍一層 ZrO 2 當溫度在 1800K, 外加電壓為 4~8kV, 使得電位能障降低, 電子很容易以熱能克服電位能陣, 此即 Schottky 效應, 其位能變化如圖 1.4 此種電子槍因為鎢針表面有氧化層保護, 具有電流穩定性高輝度大不易受外在之電磁場及振動的影響等優點三種熱游離電子槍的性質比較列於表 I 場發射電子槍場發射電子槍是利用很大的電場, 使得電位能陣變得很小, 電子藉由隧道效應 (Tunneling Effect) 直接脫離固體表面, 如圖 1.5 所示場發射電子槍構造如圖 1.6, 針尖由單晶的鎢絲製成, 方向為 [111] 或 [310], 針尖曲率半徑 (r) 約 100~1000 埃, 只要加上一很小的電壓即可使電場高達 10 7 V/cm 表 I: 電子槍性能比較熱游離槍場發射槍電子槍型態鎢絲 LaB 6 ZrO 2 /W 熱式冷式輝度 (200kV) A/cm 2 sr do 尺寸 20μm 10μm 15nm 20nm 3-10nm 壽命 100 >200 >1000 >1000 >1000 操作溫度 ( 小時 ) RT 真空 (Pa) 功函數 (ev) (100) 4.35(310) 能量分佈 (ev) 電磁透鏡電磁透鏡 (Electromagnetic Lens) 的功用, 是聚集電子束而產生放大作用其基本構造如圖 1.7 基本上, 電磁透鏡是由線圈纏繞在光軸 (Optical Axis) 周圍而組成, 這些線圈可形成均

3 勻且對稱的磁場實際上的應用, 磁場必須由軛狀鐵 (Iron Yoke) 和極片 (Pole Piece) 加以控制, 以使得電磁透鏡的作用與薄鏡 (Thin Lens) 相同偏離光軸的電子在電磁透鏡中的路徑為螺旋狀, 如圖 1.8 當電子沿著光軸行進, 如果電子正好在光軸上 ( 路徑為 A.A), 則不會受磁場影響而偏離原來的行進方向然而, 偏離光軸的電子, 在同時垂直於電子速度 v 和磁場 B 的方向上, 受力為 e(b*v), 力的大小是 evbsinθ 因此電子進入磁場後即受力 e(b*v) 而被拉向光軸, 因而形成收斂的現象, 如圖 1.8(c) 實際上電子的路徑是呈螺旋狀, 如圖 1.8(b), 因此影像不但會翻轉而且也會以光軸旋轉某一角度而呈像在螢幕上 1.4 透鏡像差透鏡像差 (Lens Aberrations) 在電子顯微鏡和光學顯微鏡均是相當重要的問題光學顯微鏡可藉由透鏡組合的設計而達到校正, 然而電子額微鏡是無法以透鏡組合的方法來校正以下依序介紹球體像差 (Spherical Aberration) 色像差 (Chromatic Aberration) 散光 (Astigmatism) 和扭曲 (Distortion) 球體像差球體像差為透鏡中最主要的缺陷, 不易校正其成因在於透鏡各部位之折射率不同, 如圖 1.9(a) 所示, 電子自光軸 P 點離開, 分別聚焦於 P' 點和 P" 點, 在影像平面 (Image Plane) 上形成一個圓盤狀影像, 其半徑為 Δr s,c s 為球體像差常數,β 為電子行進方向與光軸間之角度公式為 : Δr s =C s β 3 所有的透鏡均無法避兔球體像差, 但可使用球體像差常數較小之透鏡來降低此效應色像差色像差的成因在於不同波長的電子通過透鏡後聚焦在光軸上的不同位置, 如圖 1.9(b) 所示 Δr c 為影像半徑,C c 透鏡色像差常數,ΔE 是能量損失, 則公式為 : Δr c =C c β(δe/e) 色像差是由電子能量不同所造成, 最好的辦法是用單一波長之電子但是, 電子撞擊試片損失能量而有能量散布 (Energy spread), 待通過物鏡和投影透鏡時即又產生色像差因此, 色像差亦無法避免可採用較薄之試片或是提高加速電壓來校正散光散光的成因是透鏡具有不同的焦距 ( 此為物鏡磁場不對稱造成 ), 與電子的路徑平面 (plane of ray paths) 有關, 如圖 1.9(c) 所示兩組電子束 A B 分別走不同的路徑, 而各自聚焦於 P A P B 由於焦點不同, 形成了圓盤狀影像, 其半徑為 Δr A,Δf A 為最大焦距差關係式如下 : Δr A =Δf A β 散光可用像差補償器產生與散光像差大小相同方向相反的像差校正目前之電子顯微鏡其聚光透鏡及物鏡各有一組散光像差補償器

4 1.4.4 扭曲扭曲也是因透鏡不完美所造成, 常見的扭曲影像如圖 1.10(b) 的酒桶形 (Barrel) 扭曲和圖 1.10(c) 的針墊形 (Pincushion) 扭曲解析度解析度 (Resolution) 為某物體上之兩點經過透鏡成像後, 在可以清楚的分辨兩點之影像的條件下, 兩點的最小距離即為解析度根據 Rayleigh 準則 (Rayleigh Criterion): Δr d =0.61λ/β Δr d 為繞射像差 (Diffraction Aberration), 即解析度 λ 為電子波長圖 1.11 說明影像解析情形圖 1.11(a) 為 S 1 S 2 兩點形成繞射影像之過程圖 1.11(b), 當兩點相距夠遠時, 即可形成可解析影像圖 1.11(c), 影像正好為可解析, 此時影像已有重疊現象圖 1.11(d), 兩點間距離小於解析度, 而影像重疊圖 1.11(e),Rayleigh 準則在影像正好為可解析時, 說明影像中央的繞射強度為最大值的 0.81 倍繞射像差和透鏡本身的球體像差色像差散光對解析度均有相當程度的不良影響欲提升解析度, 可由減小球體像差常數和電子波長著手圖 1.1: 電子顯微鏡基本結構

8 二 TEM 試片製備優良的試片製備為產生優良影像的前提,TEM 試片為使電子束順利穿透, 其厚度需加以適當地減薄, 減薄的程度與加速電壓解析度需求材料種類及所感興趣的特質等有關試片製備可分為二階段, 第一階段先製成直徑 3 毫米厚約 100 微米的薄片, 第二階段則將其減薄至電子束可穿透的程度 2.1 製成直徑 3 毫米厚約 100 微米的薄片依材料來源不同, 而有不同的程序, 茲分類如下 : 延性塊材延性材料之製作可分為以下兩種方式 : (1) 以鑽石砂輪切割並配合研磨, 產生 100 微米厚的薄片, 再以打孔器或超音波切割製成 3 毫米直徑圓盤 (2) 以滾輒的方式產生直徑 3 毫米圓桿, 再以鑽石砂輪或放電加工切割機將其切成 100 微米厚或可直接將其滾輒至 100 微米厚薄片, 使用打孔器或超音波切割製成 3 毫米直徑圓盤但以上方式將導入應變或在加工期間發生熱處理現象而改變試片既有的結構脆性塊材脆性塊材之製作可分以下二方式 : (1) 如同 2.1.1(1) (2) 將材料粉碎, 並將其散佈於支撐的薄膜上, 如碳膜等非塊材依材料形態可分為以下二類 : (1) 顆粒粉末纖維等 : 1. 若厚度小於 200 奈米, 則直接散佈在支撐的膜上 2. 若厚度超過 200 奈米, 則將其鑲埋於類似的材質中形成複合物塊材, 再依塊材的處理方式製成薄片 (2) 薄膜類將其沉積於可移除的基材上, 如 : 1. 醋酸酯膜 (Acelate film) 可以利用丙銅蒸氣去除 2. 結晶鹽片可以溶於水中 3. 金屬基材, 如銅可以溶於稀硝酸中 2.2 減薄前置作業 : 把先前所得到的直徑 3 毫米厚約 100 微米薄片以凹型研磨機 (Dimpling grinder) 將薄片中央部份研磨至 3~5 微米, 再進一步減薄現有各種減薄技巧, 主要為下列四種 : 1. 視窗技術 (Window Technique) 2. 電解拋光 (Electro polishing)

9 3. 化拳拋光 (Chemical polishing) 4. 堆子噴洩 (Ion sputtering milling) 以上各技巧簡述如下 : 視窗技術 ( 廣泛使用於金屬與合金 ) 將厚 0.1~0.5 毫米一公分見方的試片以躡子夾住或點焊在白金線上, 除了試片上欲減薄的部份外, 其它部份皆塗以耐酸塗料, 試片接正極, 負極則接於具有較大表面積的白金 / 不鏽鋼管上, 按著將其浸入溶液中 ; 溶液保持攪拌並加熱至特定溫度拋光進行時維持固定溫度與外加電壓, 接著試片上方會先被貫穿, 此時反轉試片, 重新浸泡等到二端貫穿處接觸時, 即可取出試片加以清洗, 並切下貫穿處的試片加以觀察電解拋光 ( 適用於導體試片 ) 圖 2.1 顯示, 當一表面起伏的試片進行拋光時, 較突出的部份離電極較近, 電位差較大, 因而有較高的拋光速率, 經一段時間後, 試片的表面起伏被消除, 而達到拋光的效果由電解拋光原理衍生出許多拋光技術, 較常用者介紹如下 : a. 交流電噴射減薄技術 (A. C. Jet Thinning Technique) 其構造如圖 2.2 所示,Cell 1 與 Cell 2 各裝有電解液, 電解液由幫浦抽出, 經噴嘴噴向試片在噴嘴處外接一個交流電源供應器, 隨時改變電流方向, 使試片兩面能同時得到拋光效果此法可廣範地使用於陶瓷與金屬材料, 且減薄時可保留楔形凹入表面, 因此當試片被貫穿後無須立刻關機 b. 噴射缺刻 (Jet-Indenting) 其構造如圖 2.3 所示, 此法通常使用於較厚的試片 ( 約 0.2~I.0 毫米 ) 作為初步的減薄由幫浦抽出電解液, 從上方噴向試片, 控制噴嘴口徑及噴嘴距試片的距離, 可得到一平底的碟狀試片由此法得到的試片須經進一步減薄化學拋光化學拋光對於非導電性材料特別有效, 其可以運用在視窗技術與電解拋光上, 而不需要使用電極離子噴濺法離子噴濺法可用以減薄脆性或非導電性材料試片其構造如圖 2.4 所示離子在 3~6KV 的電壓加速下衝撞試片, 而將試片上的原子或分子打出減薄的速率和離子與試片原子的相對質量離子能量試片的晶體構造離子束入射角度等有關氬由於價格合理, 且是能形成離子的鈍氣中質量最重的, 因此被普遍使用在減薄的初步階段應選擇具有最大減薄率的離子入射角, 但稍後應減緩減薄率, 以減小試片受到的損害此外, 在化學拋光或電解拋光時所產生的氧化物, 方可用短暫的離子束減薄來加以消除離子噴濺法易在試片表面誘生孔洞差排, 且對於雙相材料, 可能產生不平整的表面, 此為其缺點 2.3 試片表面複製 (Replicas) 當以下情況發生時, 試片表面有必要加以複製 :

10 1. 需要較高解析度時 ( 小於 100 埃 ); 2. 試片容易為電子束所破壞 : 3. 需抽取顆粒加以識別以下介紹各種複製技巧 : 一段複製法此法適用於複製較平滑的表面原理如圖 2.5 所示, 在 10-4 mmhg 壓力下以直流電弧將碳棒蒸發, 沉積於試片上當沉積的厚度介於 100~200 埃之間時, 即可停止蒸鍍隨後將試片傾斜浸入蒸餾水中, 則碳膜複製表面將浮於水上, 與試片分離, 如圖 2.6 所示二段複製法此法適用於複製較粗糙的表面先在試片表面滴附丙酮, 再小心地放下醋酸纖維素 ( 可溶於丙酮 ) 在十分鏈內表面形成醋酸酯薄膜, 隨後將其取下, 在原先接觸試片的那一面鍍上 100~200 埃厚的碳膜後, 置於氯仿 (Chloroform) 中溶去高分子膜, 留下碳膜, 經水洗乾燥後即可使用於觀察萃取複製 (Extraction Replicas) 此法常應用於識別或計算金屬陶瓷或玻璃系統中的析出物首先將試片表面加以腐蝕, 以顯露顆粒按著以前述的一段複製法在試片表面沉積一層碳膜最後再進行腐蝕, 將碳膜連同顆粒一起剝離試片表面圖 2.1: 電解拋光機構圖圖 2.2: 交流電噴射減薄技術原理略圖

12 三操作方法本實驗依 Hitachi H-800 TEM 系統擬定操作步驟 3.1 開機 (Starting Operation) (1) Run the cooling. (2) Turn on the EVAC switch. Check if the COLUMN, GUN and CAMERA EVAC/AIR switches are at EVAC position. A high vacuum is attained in 20 to 30 min. 3.2 高壓或加速電壓使用 (Application of High Voltage or Accelerating Voltage) (1) Turn on the COL switch. (2) Check if each EVAC position of EVAC STATE GUN, COL and CAMERA lamps is lit in green. This state indicates that high vacuum is attained. (3) Depress the READY/OFF button on the left main panel and make sure that the button lamp is lit. (4) Depress any of the accelerating voltage selector switches. Voltage is gradually increased up to the selected value. (5) Check the HV/BEAM meter reading and see if the accelerating voltage is properly applied. For the initial start of the instrument, the accelerating voltage may be switched off automatically due to transitional overcurrent. If turned off, depress the READY/OFF button and apply it once again by operating the same selector switch. (6) Turn the FILAMENT control knob clockwise gradually while watching the HV/BEAM meter. Stop turning when the beam current reading is saturated on the meter. Set the filament lock at this position. (7) Manipulate the BIAS control knob so that the beam current may be saturated at 10~20μA for normal applications. 3.3 電子槍校正 (Electron Gun Alignment) (1) Set the magnification at 8,000x with the MAG control keys. (2) Depress keys 1 and 2 on the ten-key control panel. (3) Depress the FUNCTION key. CRT displays the operating procedures step by step when the DATA SET key is depressed. Follow the instructions given below. (4) Locate the beam spot at the center of the fluorescent screen by manipulating the BRIGHTNESS and BRIGHTNESS CENTERING controls. (5) Form the brightest beam spot on the fluorescent screen by manipulating the GUN TILT and GUN HORIZ controls. It does not matter even if the beam spot is off the center of the screen. (6) Bring the beam spot on the center of the screen by the GUN HORIZ controls. From a doughnut-like beam spot by under-saturating the filament using the FILAMENT control. (7) Depress the DATA SET key. 3.4 調整光束光點尺寸 (Changing the Beam Spot Size) (1) Depress key 1 on the ten-key control panel. (2) Depress the FUNCTION key. (3) Selectable beam spot sizes are displayed on CRT. Enter a desired spot size through the ten-key control panel. If you want a spot size of 3μm, just enter 3 in this order. (4) Depress the DATA SET key. (5) Depress the FUNCTION key.

13 3.5 設定底片數目 (Setting the Number of Loaded Films) (1) Depress key 2 on the ten-key control panel. (2) Depress the FUNCTION key. (3) Enter the number of loaded films through the ten-key control panel. Any number between 0 and 22 can be entered. Choose 21 sheets. (4) Depress the DATA SET key. (5) Depress the FUNCTION key. 3.6 更改使用者號碼與底片數 (Changing Operator Code and Film Number) (1) Depress key 3 on the ten-key control panel. (2) Depress the FUNCTION key. (3) Whenever depressing the DATA SET key, an asterisk * appears alternately before operator code and film number. Asterisked item is changeable. (4) Enter key 4 through the ten-key control panel, and depress the DATA SET key. (5) Depress the FUNCTION key. 3.7 設定數據 (Setting Numerical Data) (1) Depress key 4 on the ten-key control panel. (2) Depress the FUNCTION key. (3) Input numerical data from the ten-key control panel. (4) Depress the DATA SET key. An input value appears at the location for displaying film number on CRT. The value remains stored regardless of power on/off operation. In order to blank the entire data space, directly depress the DATA SET key with no numerical key depressed. (5) Depress the FUNCTION key. Input data are stored. 3.8 換試片 (Specimen Exchange) A. Specimen Loading (1) Load a specimen in the following procedures. (2) Insert the specimen holder into the slot of the specimen exchange device until it stops. Absolutely avoid turning the holder, otherwise air might be admitted into the column. (3) Turn the specimen chamber-prepumping switch toward EVAC. (4) As soon as the switch is turned on, pumping lamp (red) is lit. If the specimen holder is not completely inserted, the lamp may not be lit because the above switch does not operate. It takes about 15 seconds to complete the prepumping operation. When completed, the SPEC EVAC lamp of VACUUM STATE display is lit in green. (5) Turn the specimen holder clockwise and push it into the specimen stage. The holder may be pulled by the column vacuum. So hold the holder securely. Turn the holder must be inserted only when the SPEC EVAC lamp (green) of VACUUM STATE display is lit. B. Specimen Takeout/Unloading (1) Pull the specimen holder and turn it counterclockwise. Pull the holder once again and turn it counterclockwise. (2) Turn the specimen chamber-prepumping switch toward AIR. (3) Pull out the specimen holder. 3.9 倍率設定與對焦 (Magnification Setting and Focusing) (1) Press the MAG switch, and magnification decreases or increases. Magnification is indicated at the top of CRT. (2) Carry out focusing with the and keys of the FOCUS switch.

14 (3) Focus step is selectable between fine and coarse when depressing the FOCUS F/C switch. It is linked with magnification. When F is indicated in the FOCUS STEP on the CRT the variation rate is smaller, and when C is indicated the variation rate is large. (4) Select the field of interest and the magnification. Incline the focusing screen and carry out focusing with the FOCUS switch using the binocular. Typical images at various focusing conditions are shown below. (5) An image can easily be focused by operating the FOCUS switch while keeping the WOBBLER key depressed. In this case, the just focus is obtained when the image stops drifting. Shown below are some typical images taken at various focus conditions using the wobbler 電壓校正 (Voltage Alignment) (1) Use a uniformly distributed specimen such as microgrid and present image at 5,000 to 10,000x. (2) Depress the HV MODUL key. Radial movement is seen on the fluorescent screen image. (3) From the movement of the image, determine the voltage center (corresponding to the center of radial movement) and locate it on the center of the screen using the BEAM TILT controls. (4) If the beam spot moved off the screen, bring it back with the BRIGHTNESS CENTERING controls. (5) Turn off the HV MODUL key. (6) If the beam spot is distorted, correct it with the COND STIGM controls 電流校正 (Current Alignment) (1) Use a uniformly distributed specimen such as microgrid and present image at 5,000 to 10,000x. (2) Turn on the OBJ MODUL key. Rotating motion is seen on the fluorescent screen image. Pumping operation takes more time for a film, which has not been subjected to adequate evacuation in the plate reservoir. (11) Depress key 2 on the ten-key control panel. (12) Depress the FUNCTION key. (13) Enter the number of loaded films through the ten-key control panel. Usually it is 21 sheets. (14) Depress the DATA SET key. (15) Depress the FUNCTION key 關機 (Shutdown) (1) Turn the FILAMENT control fully counterclockwise. (2) Turn the accelerating voltage selector switch to OFF. When depressing the READY/OFF button twice, accelerating voltage turns off and the button lamp extinguishes. (3) Wait for about 5 minutes after turning off accelerating voltage, and turn off the COL switch. (4) Turn off the EVAC switch. It takes 20 minutes to cool the oil diffusion pump. (5) Turn off the main switch on the power distribution board. Close the cooling water valve.
