7 摘要摘要动脉粥样硬化 (atherosclerosis,as) 是一种严重危害人类健康的复杂血管炎性疾病, 当前针对抗动脉粥样硬化药物的研究主要通过调血脂抗炎抗氧化等方面进行有较多实验证据支持了 PPAR-α 在抗动脉粥样硬化方面的作用, 近年来研究显示大麻素受体系统与动脉粥样硬化之间有密切关系已有研究表明 PPAR-α 激动剂油酰乙醇胺 (N-oleoylethanolamine,OEA) 在调血脂方面有显著的作用本实验室前期研究显示,OEA 能够减轻 TNF-α 诱导的人脐静脉内皮细胞黏附分子表达但其作用是否单纯通过 PPAR-α 通路产生还未明确研究显示大麻素受体 2(Cannabinoid receptor 2, CB2) 在抗动脉粥样硬化中表现出一定的潜力, 抗动脉粥样硬化作用可能是由于其抗炎作用实现的本文主要研究 OEA 对人脐静脉内皮细胞 (Human Umbilical Vein Endothelial Cells,HUVEC) 抗炎相关受体大麻素受体 2 的作用本实验从新生儿脐带中提取 HUVECs 用 ELISA 法和细胞黏附实验检测 PPAR-α 阻断剂对 OEA 抗黏附的作用给予 HUVECs 不同剂量 OEA,RT-PCR 及 Western Blot 检测 CB2 基因和蛋白表达采用 PPAR-α 阻断剂 MK886 或 CB2 阻断剂 AM630 分别阻断 PPAR-α 或 CB2 信号通路, 给药组和阻断组给予 OEA, 用 TNF-α 诱导模型组给药组和阻断组炎症产生,Western Blot 法测定各分组 VCAM-1 蛋白表达或进行 THP-1 黏附实验结果表明,OEA 抗黏附的作用是部分通过 PPAR-α 通路实现的, 但不能抑制 E-selectin 表达 10 μmol L -1 OEA 和 50 μmol L -1 OEA 组的 CB2 表达上升,100μmol L -1 OEA 组较空白组无明显变化 TNF-α 诱导炎症实验中 OEA 抑制 VCAM-1 蛋白表达及 THP-1 细胞黏附, 分别给予 PPAR-α CB2 阻断剂 MK886/AM630 后,OEA 对 VCAM-1 及单核细胞黏附的抑制作用明显降低结果提示 OEA 可能通过激活 CB2 通路起到抗动脉硬化作用关键词 : 油酰乙醇胺炎症过氧化物酶体增殖物激活受体 α 大麻素受体 2 I

9 Abstract Abstract Atherosclerosis (atherosclerosis, AS) is a serious hazard to human health, current research on anti-atherosclerosis drugs mainly through aspects of lipid regulating, anti-inflammatory, anti-oxidation. Plenty of experimental evidence support the role of PPARα in the anti-atherosclerotic areas in recent years, studies have shown close relationship between the cannabinoid receptor system and atherosclerosis. Studies have shown that PPAR-α agonist oleoylethanolamide (N-oleoylethanolamine, OEA) has a significant role in regulating lipid metabolism. Our previous studies have shown, OEA can reduce TNFα-induced human umbilical vein endothelial cell adhesion molecule expression. We used healthy HUVECs and TNF-α induced HUVECs to establish a human vascular endothelial cell inflammatory model. Use ELISA and cell adhesion to assay PPARα blocker anti-adhesion effect on the OEA. To observe a PPAR-α agonist OEA s effect on CB2, an anti-inflammatory receptor in vascular endothelial cell, different dose of OEA(10,50,100 μmol L-1)had been given to HUVECs,cultured in 37 for 7 hours and then collected the total protein and total mrna, western blot detected CB2 protein expression, real-time PCR detected CB2 mrna expression. As the results show, we revealed that OEA s partial anti-adhesion effect by PPARα pathway, but can t inhibiting the expression of E-selectin.10 μmol L-1 and 50 μmol L-1 OEA could induce the CB2 protein and mrna expression, but not 100 μmol L-1.To detect if OEA anti-inflammation is partly through CB2, we used CB2 inhibitor AM630 to inhibit HUVEC CB2 expression, then detected the VCAM-1 expression induced by TNF-α, or detected THP-1 adhere to TNF-α induced HUVECs. We observed that 50 μmol L-1 OEA could inhibit TNF-α induced VCAM-1 expression and also inhibited THP-1 adhere to HUVECs, these effects could be partly inhibited by a CB2 inhibitor AM630.The effect of OEA s anti-inflammation effect is not only though PPAR-α but also though CB 2, suggesting that CB 2 signaling could be a target for II
