4 4 基 本 原 理 紅 外 線 吸 收 光 譜 法 與 紫 外 線 可 見 光 吸 收 光 譜 法 都 屬 於 分 子 吸 收 光 譜 的 範 疇 但 光 譜 產 生 的 原 理 不 同, 紅 外 線 吸 收 光 譜 是 振 動 - 轉 動 光 譜, 而 紫 外 線 可 見 光 吸 收 光 譜 是 電 子 光 譜 因 此 紅 外 線 的 光 譜 和 分 子 的 振 動 轉 動 有 密 切 關 係 紅 外 線 光 區 的 波 長 範 圍 從 μm( ,000 nm), 其 波 數 從 12,800~10 cm -1 從 儀 器 的 應 用 範 圍, 一 般 將 紅 外 線 光 區 分 為 三 個 區 : 近 紅 外 光 區 中 紅 外 光 區 及 遠 紅 外 光 區, 其 波 長 波 數 及 頻 率 範 圍 如 表 8-1 所 示 IR 儀 器 主 要 使 用 中 紅 外 光 區

5 5 一 紅 外 線 光 譜 的 基 本 概 念 帶 有 極 性 ( 偶 極 矩 ) 的 分 子 發 生 振 動 (vibration( vibration) 就 會 產 生 紅 外 線 ; 反 之, 帶 有 極 性 的 分 子 吸 收 了 與 其 振 動 能 階 對 應 的 紅 外 線 就 會 發 生 振 動 若 分 子 本 身 沒 有 極 性 ( 偶 極 矩 ), 那 就 無 法 產 生 紅 外 線 產 生 紅 外 吸 收 的 分 子 稱 為 紅 外 活 性 分 子 (IR-active), 如 CO 2 分 子 ; 反 之 為 非 紅 外 活 性 分 子 (IR-inactive), 如 O 2 分 子 A polar bond is usually IR-active A nonpolar bond in a symmetrical molecule will absorb weakly or not at all (IR-inactive)

6 Infrared Radiation Infrared Spectroscopy (IR) That part of the electromagnetic spectrum between the visible and microwave regions 0.8 μm (12,500 cm -1 ) to 50 μm (200 cm -1 ). Area of Interest in Infrared Spectroscopy The Vibrational portion of infrared spectrum 2.5 μm (4,000 cm -1) to 25 μm (400 cm -1 ) Radiation in the Vibrational Infrared region is expressed in units called wave numbers ( )( 波 數 ) Wave numbers are expressed in units of reciprocal centimeters (cm -1 ) i.e. the reciprocal of the wavelength (λ) expressed in centimeters. ν ν (cm -1 ) = 1 / λ (cm)

19 19 1. 峰 位 二 判 讀 紅 外 光 譜 的 三 要 素 分 子 內 各 種 官 能 團 的 特 徵 吸 收 峰 只 出 現 在 紅 外 光 波 譜 的 一 定 範 圍, 如 :C=O 的 伸 縮 振 動 一 般 在 1700 cm -1 左 右 2. 峰 強 紅 外 吸 收 峰 的 強 度 取 決 於 分 子 振 動 時 偶 極 矩 的 變 化, 振 動 時 分 子 偶 極 矩 的 變 化 越 小, 譜 帶 強 度 也 就 越 弱 一 般 說 來, 極 性 較 強 的 基 團 ( 如 C=O,C-X) 振 動, 吸 收 強 度 較 大 ; 極 性 較 弱 的 基 團 ( 如 C=C,N-C 等 ) 振 動, 吸 收 強 度 較 弱 ; 紅 外 吸 收 強 度 分 別 用 很 強 (vs) 強 (s) 中 (m) 弱 (w) 表 示. 3. 峰 形 不 同 基 團 的 某 一 種 振 動 形 式 可 能 會 在 同 一 頻 率 範 圍 內 都 有 紅 外 吸 收, 如 -OH -NH 的 伸 縮 振 動 峰 都 在 cm -1 但 二 者 峰 形 狀 有 顯 著 不 同 此 時 峰 形 的 不 同 有 助 於 官 能 團 的 鑒 別

20 20 分 子 鍵 結 與 紅 外 線 吸 收 之 關 係 1. 依 虎 克 定 律 的 解 說 可 瞭 解, 兩 個 物 質 間 的 振 動 頻 率 與 兩 個 物 質 的 質 量 及 兩 個 物 質 間 的 作 用 力 有 關 ; 一 般 而 言, 質 量 越 大 振 動 越 慢 ; 作 用 力 越 大 振 動 越 快, 但 不 管 如 何 振 動 越 快 就 代 表 所 需 的 能 量 越 高 2. 將 依 虎 克 定 律 用 在 分 子 模 型 時, 也 可 用 來 解 釋 分 子 鍵 結 間 振 動 的 模 式 : 兩 個 原 子 鍵 結 的 振 動 頻 率 與 兩 個 原 子 的 質 量 及 兩 個 原 子 間 的 鍵 結 力 量 有 關 ; 一 般 而 言, 原 子 的 質 量 越 大 振 動 越 慢 ; 但 鍵 結 力 越 大 則 振 動 越 快, 而 振 動 越 快 就 代 表 所 吸 收 或 發 射 的 波 長 越 短 3. 依 原 子 質 量 而 言 : 振 動 頻 率 C-H > C-N > C-O > C-Br 4. 依 鍵 結 力 量 而 言 : 振 動 頻 率 C H > C=H > C-H 5. 依 振 動 的 方 式 而 言 : 振 動 頻 率 為 伸 縮 振 動 > 彎 曲 振 動

24 三 紅 外 光 譜 的 應 用 紅 外 光 譜 的 最 大 特 點 是 具 有 特 徵 性, 譜 圖 上 的 每 個 吸 收 峰 代 表 了 分 子 中 某 個 基 團 的 特 定 振 動 形 式 據 此 進 行 化 合 物 的 定 性 分 析 和 定 量 分 析 廣 泛 應 用 於 石 油 化 工 生 物 醫 藥 環 境 監 測 等 方 面 1. 定 性 分 析 (1) 已 知 物 的 鑒 定 在 得 到 試 樣 的 紅 外 譜 圖 後, 與 純 物 質 的 譜 圖 進 行 比 較, 如 果 譜 圖 中 峰 位 峰 形 和 峰 的 相 對 強 度 都 一 致, 即 可 認 為 是 同 一 物 質 (2) 未 知 物 的 鑒 定 是 紅 外 光 譜 法 定 性 分 析 的 一 個 重 要 用 途, 涉 及 到 圖 譜 的 解 析 首 先 應 瞭 解 樣 品 的 來 源 用 途 製 備 方 法 分 離 方 法 理 化 性 質 元 素 組 成 及 其 它 光 譜 分 析 資 料 如 UV NMR MS 等 有 助 於 對 樣 品 結 構 資 訊 的 歸 屬 和 辨 認 24

25 25 四 紅 外 光 譜 的 應 用 下 面 列 舉 兩 例 加 以 說 明 : 例 一 : 化 合 物 C 8 H 8 O 2 的 紅 外 光 譜 如 右 圖 所 示, 試 推 測 其 結 構 Ar-H -CH 解 : 計 算 不 飽 和 度 : Ω = + + n n n Ω = + 2 = 5 -CH 3 υ C=O C=C -CH 3 γ CH 苯 C H 3000 cm-1 有 吸 收, 說 明 有 和 ==C-H 基 團 存 在 靠 近 1700 cm-1 的 強 度 吸 收, 表 O 明 有 C=O 基 團, 結 合 2730 cm-1 特 徵 峰, 進 一 步 說 明 有 醛 基 存 在, C H cm-1 有 吸 收 說 明 有 苯 環 存 在 根 據 820 cm-1 吸 收 帶 苯 為 對 位 取 代 cm-1 是 -CH3 特 徵 吸 收 峰, 根 據 以 上 解 析 並 對 照 標 準 譜 圖 確 定 化 合 物 為 茴 香 醛 : H 3 CO CHO

26 26 四 紅 外 光 譜 的 應 用 例 二 : 下 列 反 應 可 能 生 成 (A) 和 (B) 兩 個 異 構 體, 用 1H-NMR 分 析, 化 學 位 移 都 一 樣 ; 質 譜 MS 分 子 離 子 峰 也 一 樣 用 紅 外 光 譜 觀 測 時, 在 1775 cm-1 附 近 只 看 到 一 個 吸 收 峰, 由 此 確 定 反 應 生 成 物 為 B 若 為 A, 屬 酸 酐 類 化 合 物?C=O 應 有 兩 個 吸 收 峰 (A) (B)

27 27 2. 定 量 分 析 三 紅 外 光 譜 的 應 用 定 量 依 據 是 Lambert- Beer 定 律, 定 量 時 吸 光 度 的 測 定 常 用 基 線 法 如 圖 所 示, 圖 中 I 與 I0 之 比 就 是 透 射 比 基 線 的 畫 法

28 1. 對 試 樣 的 要 求 四 樣 品 製 備 技 術 (1) 試 樣 應 該 是 單 一 組 分 的 純 物 質, 純 度 應 >98%, 便 於 與 純 化 合 物 的 標 準 進 行 對 照 多 組 分 試 樣 應 在 測 定 前 儘 量 預 先 用 分 餾 萃 取 重 結 晶 區 域 熔 融 或 色 譜 法 進 行 分 離 提 純 (2) 試 樣 中 不 應 含 有 游 離 水 水 本 身 有 紅 外 吸 收, 會 嚴 重 干 擾 樣 品 譜, 而 且 還 會 侵 蝕 吸 收 池 的 鹽 窗 (3) 試 樣 的 濃 度 和 測 試 厚 度 應 選 擇 適 當, 以 使 光 譜 圖 中 的 大 多 數 吸 收 峰 的 透 射 度 處 於 15%~70% 範 圍 內 (4) 直 接 測 定 固 體 樣 品 時, 要 求 樣 品 顆 粒 直 徑 小 於 紅 外 線 之 波 長, 否 則 會 發 生 對 入 射 光 明 顯 的 散 射 28

29 29 2. 樣 品 的 製 備 四 樣 品 製 備 技 術 (1) 固 體 樣 品 的 製 備 a. 壓 片 法 : 將 1~2mg 固 體 試 樣 與 200mg 純 KBr 研 細 混 合, 研 磨 到 粒 度 小 於 2μm, 在 油 壓 機 上 壓 成 透 明 薄 片, 即 可 用 於 測 定 b. 糊 狀 法 : 研 細 的 固 體 粉 末 和 石 蠟 油 調 成 糊 狀, 塗 在 兩 片 空 白 之 氯 化 鈉 窗 片 上, 進 行 測 試 此 法 可 消 除 水 峰 的 干 擾 液 體 石 蠟 本 身 有 紅 外 吸 收, 此 法 不 能 用 來 研 究 飽 和 烷 烴 的 紅 外 吸 收

30 30 四 樣 品 製 備 技 術 薄 膜 法 : a. 熔 融 法 : 對 熔 點 低, 在 熔 融 時 不 發 生 分 解 昇 華 和 其 他 化 學 變 化 的 物 質, 用 熔 融 法 製 備 可 將 樣 品 直 接 用 紅 外 燈 或 電 吹 風 加 熱 熔 融 後 塗 製 成 膜 b. 熱 壓 成 膜 法 : 對 於 某 些 聚 合 物 可 把 它 們 放 在 兩 塊 具 有 拋 光 面 的 金 屬 塊 間 加 熱, 樣 品 熔 融 後 立 即 用 油 壓 機 加 壓, 冷 卻 後 揭 下 薄 膜 夾 在 夾 具 中 直 接 測 試 c. 溶 液 制 膜 法 : 將 試 樣 溶 解 在 低 沸 點 的 易 揮 發 溶 劑 中, 塗 在 鹽 片 上, 待 溶 劑 揮 發 後 成 膜 來 測 定 如 果 溶 劑 和 樣 品 不 溶 于 水, 使 它 們 在 水 面 上 成 膜 也 是 可 行 的 比 水 重 的 溶 劑 在 汞 表 面 成 膜

31 31 四 樣 品 製 備 技 術 (2) 液 體 樣 品 的 製 備 a. 液 膜 法 : 對 沸 點 較 高 的 液 體, 直 接 滴 在 兩 塊 鹽 片 之 間, 形 成 沒 有 氣 泡 的 毛 細 厚 度 液 膜, 然 後 用 夾 具 固 定, 放 入 儀 器 光 路 中 進 行 測 試 b. 液 體 吸 收 槽 法 : 對 於 低 沸 點 液 體 樣 品 和 定 量 分 析, 要 用 固 定 密 封 液 體 池 制 樣 時 液 體 池 傾 斜 放 置, 樣 品 從 下 口 注 入, 直 至 液 體 被 充 滿 為 止, 用 聚 四 氟 乙 烯 塞 子 依 次 堵 塞 池 的 入 口 和 出 口, 進 行 測 試 (3) 氣 態 樣 品 的 製 備 : 氣 態 樣 品 一 般 都 灌 注 于 氣 體 池 內 進 行 測 試

33 33 光 源 Infrared Sources-1- 能 士 特 發 光 體 The Nernst Glower or Nernst Lamp. An early light bulb which did not need to operate in vacuum like Edison s W filament bulb. A ZrO 2 :Y 2 O 3 :Er 2 O 3 (90:7:3) mixed oxide. A current passing through it would heat it to glowing. Needed an external filament to get it hot to begin, but then it could continue on its own. Gives a good spectrum from 1 to 10 µm; intensity varies 3 orders of magnitude

34 34 Infrared Sources 2- 熾 棒 光 源 The Globar is a competitor of the Nernst Glower. Similar concept, but uses SiC instead of metal oxides. Can start heating from room temperature. Globar is a bit better for wavelengths below 5 µm; also the Globar requires water cooling of the filament supports.

35 35 Infrared Sources 3- 白 熱 燈 絲 光 源 Incandescent wire sources are longer lasting but lower intensity than the glower or globar. 78 Chem 315 Lab Experiment Overview Notes 78 Infrared Spectroscopy (IR) No two molecules of different structure will have exactly the same natural frequency of vibration, each will have a unique infrared absorption pattern or spectrum. Two Uses: 1. IR can be used to distinguish one compound from another. 2. Absorption of IR energy by organic compounds will occur in a manner characteristic of the types of bonds and atoms in the functional groups present in the compound; thus, infrared spectrum gives structural information about a molecule. The absorptions of each type of bond (N H, C H, OH, C X, C=O, C O, C C, C=C, C C, C N, etc.) are regularly found only in certain small portions of the vibrational infrared region, greatly enhancing analysis possibilities.

84 Chem 315 Lab Experiment Overview Notes Step 3 (Con t) Aromatic Ring Absorptions Infrared Spectroscopy (IR) Aromatic unsaturated C=C bonds show an absorption on the left side of 3000 cm -1, but the aromaticity must be verified in the overtone region ( cm -1 ) and the out-of-plane (OOP) region ( cm -1 ) 4 Medium to strong absorptions in region cm -1 Many weak combination and overtone absorptions appear between 2000 and 1667 cm -1 The relative shapes and numbers (1-4) of the overtone absorptions can be used to tell whether the aromatic ring is monosubstituted or di-, tri-, tetra-, penta-, or hexasubstituted. Positional (ortho (o), meta (m), para (p)) isomers can also be distinguished. Note: A strong carbonyl absorption can overlap these overtone bands, making them unusable. 84

The number of absorptions and their relative positions are unique to each type of substitution. Although these absorptions are in the Fingerprint region they are particularly reliable for rings with Alkyl group substitutions. They are less reliable for Polar substituents.
